Abstract
UA : Дана дипломна робота обсягом 84 сторінки друкованого тесту, яка містить 25 рисунків та 7 таблиць. Під час виконання роботи було опрацьовано 74 наукових джерела, з яких 36 є англомовними джерелами.
Об’єктом дослідження стали сучасні методики виділення та аналізу ДНК різних генотипів льону олійного.
Льон відноситься до найдавніших культур, які використовуються людиною. Щороку він має великі посівні площі в багатьох країнах на різних континентах. Але незважаючи на поширеність та важливе економічне значення цієї культури, генетика льону вивчена недостатньо. Застосування сучасних молекулярно-генетичних методів для дослідження структури і мінливості генетичного апарату цієї цінної культури є дуже актуальним напрямом. Дослідження на молекулярному рівні дозволять розкрити генетичні особливості льону, що сприятиме підвищенню його продуктивності і конкурентоспроможності.
Метою даної роботи було проведення молекулярно-генетичного аналізу мутантів льону олійного для створення ДНК-бібліотеки і поглиблення рівня генетичної роботи з цією цінною культурою.
У ході виконання експерементальної частини кваліфікаційної роботи застосовувалися наступні методи молекулярних досліджень: виділення ДНК методом MagMaxTM Plant DNA Izolation Kit, електрофорез отриманих молекул ДНК, спектрофотометрія та створення бібліотеки льону олійного з використанням набору KAPA HyperPlus Kit.
В результаті проведеної роботи було успішно освоєно методики молекулярно-генетичних досліджень, зокрема виділення ДНК методом MagMaxTM Plant DNA Izolation Kit та створення ДНК-бібліотеки льону олійного. Найвищу концентрацію ДНК отримано з молодих листків. Застосування KAPA HyperPlus Kit дозволило ефективно здійснити фрагментацію та ампліфікацію ДНК, що значно скоротило час підготовки зразків. Використання високочутливого D1000 DNA ScreenTape забезпечило точний аналіз ДНК-бібліотеки 17 досліджуваних генотипів. Результати підтвердили ефективність обраних методів для подалього дослідження геному льону олійного.
EN : The thesis consists of 84 printed pages featuring 25 figures and seven tables. During its preparation, 74 scientific sources were analysed, 36 of which were English-language references. The study focuses on modern methods for extracting and analysing DNA from various genotypes of oil flax. Flax is among the oldest crops utilised by humans and is cultivated extensively across continents, occupying significant sowing areas in numerous countries annually. Despite its widespread cultivation and economic importance, the genetics of flax remains insufficiently studied. Applying modern molecular-genetic methods to investigate the structure and variability of the genetic apparatus of this valuable crop is a highly relevant research direction. Molecular-level studies will uncover the genetic characteristics of flax, enhancing its productivity and competitiveness. This research aimed to perform a molecular-genetic analysis of oil flax mutants, create a DNA library, and deepen the genetic study of this valuable crop. Methods and Experimental Procedures The experimental section employed the following molecular research methods: DNA extraction using the MagMax™ Plant DNA Isolation Kit, Electrophoresis of extracted DNA molecules, Spectrophotometry, and Creation of a DNA library for oil flax using the KAPA HyperPlus Kit. The research successfully mastered molecular-genetic methods, including DNA extraction with the MagMax™ Plant DNA Isolation Kit and creating an oil flax DNA library. The highest DNA concentrations were obtained from young leaves. The KAPA HyperPlus Kit was effectively used for DNA fragmentation and amplification, significantly reducing sample preparation time. High-sensitivity D1000 DNA ScreenTape ensured accurate analysis of the DNA library for 17 studied genotypes. The results confirmed the efficiency of the selected methods for further genome studies of oil flax.
