Abstract
UA : В роботі 88 сторінок, 1 таблиця, 24 рисунки, використано 77 літературних джерел, із них 61 іноземною мовою.
Об’єктом дослідження є мутантні форми соняшника (Helianthus annuus L.) – вихідна лінія Zl95 та її мутанти зі зміненим габітусом: "Fanleaf" (віялове жилкування), "Brown-red" (червоно-коричневе насіння) та "Tobacco" (карликовий, тютюноподібний тип).
Предметом дослідження є молекулярно-генетичні зміни, асоційовані із зміненим фенотипом у мутантних форм соняшника.
Методи досліджень включали біоінформатичний аналіз транскриптомних даних, класичні молекулярно-біологічні методи, метод секвенування за Сенгером та подальший біоінформатичний аналіз.
Метою кваліфікаційної роботи є молекулярно-генетичний аналіз мутантних форм соняшника із зміненим габітусом, отриманих за допомогою хімічного мутагенезу ЕМС, для ідентифікації ключових генетичних змін, що обумовлюють їх фенотипові відмінності.
Теоретично та експериментально визначено:
1. Ефективність хімічного мутагенезу ЕМС для створення широкого спектру мутантних форм соняшника зі зміненою морфологією.
2. Специфічні транскриптомні профілі кожної мутантної форми: "Brown-red" – інтенсивна активація шляхів синтезу білка та фотосинтезу; "Fanleaf" – активація генів захисту та метаболізму вторинних сполук (терпени); "Tobacco" – проміжний профіль із акцентом на базові клітинні процеси.
3. Ключові диференційно експресовані гени-кандидати, серед яких особливу увагу привернув ген терпенсинтази TPS10.
4. Експериментальне підтвердження змін експресії гена TPS10 у мутанті "Brown-red" методом qPCR.
5. Наявність конкретних точкових мутацій (переходи типу G→A, C→T, характерні для EMS), вставок та випадінь у гені TPS10 у досліджуваних мутантних лініях, що є потенційно каузативними для змінених фенотипів.
6. Практичну цінність отриманих результатів для селекції, що полягає у можливості розробки молекулярних маркерів для цілеспрямованого відбору форм соняшника з оптимальним габітусом та покращеними адаптивними властивостями.
EN : The thesis comprises 88 pages, 1 tables, 24 figures, and references 77 literature sources, of which 61 are in foreign languages. The object of the research is mutant forms of sunflower (Helianthus annuus L.) – the initial line Zl95 and its mutants with altered habitus: "Fanleaf" (fan-shaped venation), "Brown-red" (Brown-red seed color), and "Tobacco" (dwarf, tobacco-like type). The subject of the research is the molecular-genetic changes associated with the altered phenotype in the sunflower mutant forms. Research methods included bioinformatic analysis of transcriptomic data, classical molecular-biological methods, Sanger sequencing, and subsequent bioinformatic analysis. The aim of the qualification work is the molecular-genetic analysis of sunflower mutants with altered habitus, obtained through EMS chemical mutagenesis, to identify key genetic changes that determine their phenotypic differences. The following was determined theoretically and experimentally: 1. The effectiveness of EMS chemical mutagenesis for creating a wide spectrum of sunflower mutant forms with altered morphology. 2. Specific transcriptomic profiles for each mutant form: "Brown-red" – intense activation of protein synthesis and photosynthesis pathways; "Fanleaf" – activation of defense genes and metabolism of secondary compounds (terpenes); "Tobacco" – an intermediate profile with an emphasis on basic cellular processes. 3. Key differentially expressed candidate genes, among which the terpene synthase gene TPS10 attracted particular attention. 4. Experimental confirmation of changes in TPS10 gene expression in the "Brown-red" mutant using qPCR. 5. The presence of specific point mutations (G→A, C→T transitions, characteristic of EMS) and indels in the TPS10 gene in the studied mutant lines, which are potentially causative for the altered phenotypes. 6. The practical value of the obtained results for breeding, which lies in the possibility of developing molecular markers for the targeted selection of sunflower forms with optimal habitus and improved adaptive traits.
